Uncertain significance for Leber congenital amaurosis 6; Cone-rod dystrophy 13 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020366.4(RPGRIP1):c.620G>A (p.Ser207Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with asparagine at codon 207 of the RPGRIP1 protein (p.Ser207Asn). The serine residue is weakly conserved and there is a small physicochemical difference between serine and asparagine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has not been reported in the literature in individuals affected with RPGRIP1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
