This variant lies in the MYH7 gene (transcript NM_000257.4) at coding-DNA position 1753, where A is replaced by G; at the protein level this means replaces isoleucine at residue 585 with valine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with valine at codon 585 of the MYH7 protein (p.Ile585Val). The isoleucine residue is weakly conserved and there is a small physicochemical difference between isoleucine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with MYH7-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant is found within a region of MYH7 between codons 181 and 937 that contains the majority of the myosin head domain.
