NM_004655.4(AXIN2):c.4A>G (p.Ser2Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S2G variant (also known as c.4A>G), located in coding exon 1 of the AXIN2 gene, results from an A to G substitution at nucleotide position 4. The serine at codon 2 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
