Uncertain significance for Short-rib thoracic dysplasia 8 with or without polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018051.5(DYNC2I1):c.2172G>C (p.Trp724Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tryptophan with cysteine at codon 724 of the WDR60 protein (p.Trp724Cys). The tryptophan residue is highly conserved and there is a large physicochemical difference between tryptophan and cysteine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with clinical features of short-rib thoracic dysplasia (Invitae). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
