Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.1002G>T (p.Lys334Asn), citing Ambry Variant Classification Scheme 2023: The p.K334N variant (also known as c.1002G>T), located in coding exon 4 of the MSH6 gene, results from a G to T substitution at nucleotide position 1002. The lysine at codon 334 is replaced by asparagine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
