Uncertain significance for Leber congenital amaurosis 1; Cone-rod dystrophy 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000180.4(GUCY2D):c.2015G>A (p.Cys672Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine with tyrosine at codon 672 of the GUCY2D protein (p.Cys672Tyr). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and tyrosine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with Leber congenital amaurosis (PMID: 23661368) or clinical features of inherited retinal disease (Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
