This variant lies in the SYNE1 gene (transcript NM_182961.4) at coding-DNA position 8609, where C is replaced by T; at the protein level this means replaces serine at residue 2870 with phenylalanine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with SYNE1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces serine with phenylalanine at codon 2877 of the SYNE1 protein (p.Ser2877Phe). The serine residue is highly conserved and there is a large physicochemical difference between serine and phenylalanine. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
