Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004415.4(DSP):c.7991C>A (p.Thr2664Asn), citing Ambry Variant Classification Scheme 2023: The p.T2664N variant (also known as c.7991C>A), located in coding exon 24 of the DSP gene, results from a C to A substitution at nucleotide position 7991. The threonine at codon 2664 is replaced by asparagine, an amino acid with similar properties. This alteration has been reported in an individual with left ventricular hypertrabeculations and an additional alteration in another cardiac-related gene (Miszalski-Jamka K et al. Circ Cardiovasc Genet, 2017 Aug;10:[ePub ahead of print]). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
