Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_023036.6(DNAI2):c.965C>A (p.Ser322Tyr), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DNAI2 gene (transcript NM_023036.6) at coding-DNA position 965, where C is replaced by A; at the protein level this means replaces serine at residue 322 with tyrosine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This sequence change replaces serine with tyrosine at codon 322 of the DNAI2 protein (p.Ser322Tyr). The serine residue is moderately conserved and there is a large physicochemical difference between serine and tyrosine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with DNAI2-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
