Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000393.5(COL5A2):c.4295A>G (p.Asp1432Gly), citing Ambry Variant Classification Scheme 2023: The c.4295A>G (p.D1432G) alteration is located in exon 53 (coding exon 53) of the COL5A2 gene. This alteration results from a A to G substitution at nucleotide position 4295, causing the aspartic acid (D) at amino acid position 1432 to be replaced by a glycine (G). Based on data from gnomAD, the G allele has an overall frequency of 0.003% (1/31406) total alleles studied. The highest observed frequency was 0.007% (1/15432) of European (non-Finnish) alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
