Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_177438.3(DICER1):c.1134C>G (p.Ile378Met), citing Sema4 Curation Guidelines. This variant lies in the DICER1 gene (transcript NM_177438.3) at coding-DNA position 1134, where C is replaced by G; at the protein level this means replaces isoleucine at residue 378 with methionine — a missense variant. Submitter rationale: The DICER1 c.1134C>G (p.I378M) variant has not been reported in the literature to our knowledge. It was observed in 2/34592 chromosomes of the Latino subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 1037266). Computational analyses and evolutionary conservation data do not provide strong support for or against an impact to the protein, however these predictions have not been confirmed by published functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
