Uncertain significance for DICER1-related tumor predisposition — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_177438.3(DICER1):c.4010A>C (p.Asp1337Ala), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DICER1 gene (transcript NM_177438.3) at coding-DNA position 4010, where A is replaced by C; at the protein level this means replaces aspartic acid at residue 1337 with alanine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with DICER1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces aspartic acid with alanine at codon 1337 of the DICER1 protein (p.Asp1337Ala). The aspartic acid residue is highly conserved and there is a moderate physicochemical difference between aspartic acid and alanine.
