Uncertain significance for Distal myopathy with posterior leg and anterior hand involvement; Myofibrillar myopathy 5; Hypertrophic cardiomyopathy 26 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001458.5(FLNC):c.7819T>G (p.Ser2607Ala), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the FLNC gene (transcript NM_001458.5) at coding-DNA position 7819, where T is replaced by G; at the protein level this means replaces serine at residue 2607 with alanine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with FLNC-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces serine with alanine at codon 2607 of the FLNC protein (p.Ser2607Ala). The serine residue is highly conserved and there is a moderate physicochemical difference between serine and alanine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
