Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.7418G>A (p.Gly2473Asp), citing Ambry Variant Classification Scheme 2023: The p.G2473D variant (also known as c.7418G>A), located in coding exon 59 of the FBN1 gene, results from a G to A substitution at nucleotide position 7418. The glycine at codon 2473 is replaced by aspartic acid, an amino acid with similar properties. This variant was reported in individual(s) with features consistent with aortic aneurysm or dissection and segregated with disease in at least one family (external communication; Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
