NM_198253.3(TERT):c.608G>C (p.Trp203Ser) was classified as Uncertain significance for Dyskeratosis congenita by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W203S variant (also known as c.608G>C), located in coding exon 2 of the TERT gene, results from a G to C substitution at nucleotide position 608. The tryptophan at codon 203 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
