Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000368.5(TSC1):c.1151A>G (p.Lys384Arg), citing Ambry Variant Classification Scheme 2023: The p.K384R variant (also known as c.1151A>G), located in coding exon 10 of the TSC1 gene, results from an A to G substitution at nucleotide position 1151. The lysine at codon 384 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
