This variant lies in the CASR gene (transcript NM_000388.4) at coding-DNA position 41, where C is replaced by A; at the protein level this means replaces threonine at residue 14 with asparagine — a missense variant. Submitter rationale: This sequence change replaces threonine with asparagine at codon 14 of the CASR protein (p.Thr14Asn). The threonine residue is moderately conserved and there is a small physicochemical difference between threonine and asparagine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with CASR-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
