This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 3527, where T is replaced by C; at the protein level this means replaces valine at residue 1176 with alanine — a missense variant. Submitter rationale: This sequence change replaces valine with alanine at codon 1176 of the DSP protein (p.Val1176Ala). The valine residue is moderately conserved and there is a small physicochemical difference between valine and alanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with DSP-related conditions.
