Uncertain significance for Larsen-like syndrome, B3GAT3 type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012200.4(B3GAT3):c.58CTC[1] (p.Leu21del), citing Invitae Variant Classification Sherloc (09022015): This variant, c.61_63del, results in the deletion of 1 amino acid(s) of the B3GAT3 protein (p.Leu21del), but otherwise preserves the integrity of the reading frame. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with clinical features B3GAT3-related conditions (Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
