This variant lies in the TMPO gene (transcript NM_001032283.3) at coding-DNA position 380, where A is replaced by G; at the protein level this means replaces tyrosine at residue 127 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tyrosine with cysteine at codon 127 of the TMPO protein (p.Tyr127Cys). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and cysteine.
