NM_018062.4(FANCL):c.1021T>A (p.Trp341Arg) was classified as Uncertain significance for Fanconi anemia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tryptophan with arginine at codon 341 of the FANCL protein (p.Trp341Arg). The tryptophan residue is highly conserved and there is a moderate physicochemical difference between tryptophan and arginine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with clinical features of Fanconi anemia (PMID: 26740942).
