Uncertain significance for Combined immunodeficiency due to MALT1 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006785.4(MALT1):c.133C>G (p.Leu45Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 45 of the MALT1 protein (p.Leu45Val). This variant is present in population databases (rs549192343, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with MALT1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1031677). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated.
