Pathogenic for Familial hemiplegic migraine — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000702.4(ATP1A2):c.1843G>A (p.Gly615Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 615 of the ATP1A2 protein (p.Gly615Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal dominant familial hemiplegic migraine (PMID: 16437583, 20837964). In at least one individual the variant was observed to be de novo. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 1029935). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt ATP1A2 protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects ATP1A2 function (PMID: 16437583). For these reasons, this variant has been classified as Pathogenic.
