Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002768.5(CHMP1A):c.130T>A (p.Cys44Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine, which is neutral and slightly polar, with serine, which is neutral and polar, at codon 44 of the CHMP1A protein (p.Cys44Ser). This variant is present in population databases (rs372234497, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with CHMP1A-related conditions. ClinVar contains an entry for this variant (Variation ID: 1028045). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
