Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.7276T>G (p.Cys2426Gly), citing Ambry Variant Classification Scheme 2023: The p.C2427G variant (also known as c.7279T>G), located in coding exon 8 of the ALMS1 gene, results from a T to G substitution at nucleotide position 7279. The cysteine at codon 2427 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
