Uncertain significance for Tumor predisposition syndrome 3 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015450.3(POT1):c.834G>T (p.Leu278Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with phenylalanine at codon 278 of the POT1 protein (p.Leu278Phe). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and phenylalanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with POT1-related conditions.
