This variant lies in the SPTLC2 gene (transcript NM_004863.4) at coding-DNA position 857, where A is replaced by G; at the protein level this means replaces glutamine at residue 286 with arginine — a missense variant. Submitter rationale: This sequence change replaces glutamine with arginine at codon 286 of the SPTLC2 protein (p.Gln286Arg). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and arginine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with SPTLC2-related conditions.
