NM_002661.5(PLCG2):c.1173C>A (p.Asp391Glu) was classified as Uncertain significance for Familial cold autoinflammatory syndrome 3 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The glutamic acid amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with PLCG2-related conditions. This variant is present in population databases (rs772084816, ExAC 0.006%). This sequence change replaces aspartic acid with glutamic acid at codon 391 of the PLCG2 protein (p.Asp391Glu). The aspartic acid residue is highly conserved and there is a small physicochemical difference between aspartic acid and glutamic acid.
