NM_000302.4(PLOD1):c.1165G>A (p.Glu389Lys) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E389K variant (also known as c.1165G>A), located in coding exon 11 of the PLOD1 gene, results from a G to A substitution at nucleotide position 1165. The glutamic acid at codon 389 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
