Uncertain significance for Glycine encephalopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004483.5(GCSH):c.254G>T (p.Cys85Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine with phenylalanine at codon 85 of the GCSH protein (p.Cys85Phe). The cysteine residue is moderately conserved and there is a large physicochemical difference between cysteine and phenylalanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with GCSH-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
