Uncertain significance for Emery-Dreifuss muscular dystrophy 4, autosomal dominant; Autosomal recessive ataxia, Beauce type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_182961.4(SYNE1):c.22837G>C (p.Val7613Leu), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with SYNE1-related conditions. This variant is present in population databases (rs776253447, ExAC 0.003%). This sequence change replaces valine with leucine at codon 7542 of the SYNE1 protein (p.Val7542Leu). The valine residue is highly conserved and there is a small physicochemical difference between valine and leucine.
