Uncertain significance for Glycogen storage disease due to muscle and heart glycogen synthase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002103.5(GYS1):c.2032G>A (p.Asp678Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 678 of the GYS1 protein (p.Asp678Asn). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with GYS1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1024883). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
