NM_007126.5(VCP):c.268A>G (p.Asn90Asp) was classified as Uncertain significance for Inclusion body myopathy with Paget disease of bone and frontotemporal dementia; Frontotemporal dementia and/or amyotrophic lateral sclerosis 6 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt VCP protein function. This variant has been observed in individual(s) with clinical features of amyotrophic lateral sclerosis (Invitae). This variant is not present in population databases (ExAC no frequency). This sequence change replaces asparagine with aspartic acid at codon 90 of the VCP protein (p.Asn90Asp). The asparagine residue is highly conserved and there is a small physicochemical difference between asparagine and aspartic acid.
