Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.68G>A (p.Arg23Gln), citing Ambry Variant Classification Scheme 2023: The p.R23Q variant (also known as c.68G>A), located in coding exon 1 of the AXIN2 gene, results from a G to A substitution at nucleotide position 68. The arginine at codon 23 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
