Uncertain significance for Craniolenticulosutural dysplasia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006364.4(SEC23A):c.631C>A (p.Leu211Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 211 of the SEC23A protein (p.Leu211Ile). This variant is present in population databases (rs8018720, gnomAD 0.07%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with SEC23A-related conditions. ClinVar contains an entry for this variant (Variation ID: 1024104). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign".
