NM_002485.5(NBN):c.1213G>A (p.Glu405Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E405K variant (also known as c.1213G>A), located in coding exon 10 of the NBN gene, results from a G to A substitution at nucleotide position 1213. The glutamic acid at codon 405 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species; however, lysine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
