This variant lies in the SCN11A gene (transcript NM_001349253.2) at coding-DNA position 5075, where G is replaced by T; at the protein level this means replaces arginine at residue 1692 with methionine — a missense variant. Submitter rationale: This sequence change replaces arginine with methionine at codon 1692 of the SCN11A protein (p.Arg1692Met). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and methionine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with SCN11A-related conditions.
