Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_000548.5(TSC2):c.1361+6G>A, citing Sema4 Curation Guidelines. This variant lies in the TSC2 gene (transcript NM_000548.5) at 6 bases into the intron immediately after coding-DNA position 1361, where G is replaced by A. Submitter rationale: The TSC2 c.1361+6G>A variant has not been reported in the literature to our knowledge. It was not observed in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org). The variant has been reported in ClinVar (Variation ID: 1023016). Splice site prediction tools suggest the variant does not disrupt normal splicing, however these predictions have not been confirmed by transcriptional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
