Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.2998G>C (p.Glu1000Gln), citing Ambry Variant Classification Scheme 2023: The p.E1000Q variant (also known as c.2998G>C), located in coding exon 18 of the ALK gene, results from a G to C substitution at nucleotide position 2998. The glutamic acid at codon 1000 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
