This variant lies in the NBN gene (transcript NM_002485.5) at coding-DNA position 587, where T is replaced by A; at the protein level this means replaces phenylalanine at residue 196 with tyrosine — a missense variant. Submitter rationale: This sequence change replaces phenylalanine with tyrosine at codon 196 of the NBN protein (p.Phe196Tyr). The phenylalanine residue is highly conserved and there is a small physicochemical difference between phenylalanine and tyrosine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with NBN-related conditions.
