Uncertain significance for Short-rib thoracic dysplasia 8 with or without polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018051.5(DYNC2I1):c.1505C>T (p.Ser502Leu), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DYNC2I1 gene (transcript NM_018051.5) at coding-DNA position 1505, where C is replaced by T; at the protein level this means replaces serine at residue 502 with leucine — a missense variant. Submitter rationale: This variant is present in population databases (rs776188298, ExAC 0.002%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15"). This variant has not been reported in the literature in individuals with WDR60-related disease. This sequence change replaces serine with leucine at codon 502 of the WDR60 protein (p.Ser502Leu). The serine residue is highly conserved and there is a large physicochemical difference between serine and leucine.
