Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001103.4(ACTN2):c.955A>G (p.Lys319Glu), citing Ambry Variant Classification Scheme 2023: The p.K319E variant (also known as c.955A>G), located in coding exon 10 of the ACTN2 gene, results from an A to G substitution at nucleotide position 955. The lysine at codon 319 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
