NM_017534.6(MYH2):c.762C>A (p.His254Gln) was classified as Uncertain significance for Myopathy, proximal, and ophthalmoplegia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine with glutamine at codon 254 of the MYH2 protein (p.His254Gln). The histidine residue is highly conserved and there is a small physicochemical difference between histidine and glutamine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). This variant has not been reported in the literature in individuals with MYH2-related conditions. This variant is not present in population databases (ExAC no frequency).
