Uncertain significance for Dyskeratosis congenita — the classification assigned by Ambry Genetics to NM_198253.3(TERT):c.365A>G (p.Tyr122Cys), citing Ambry Variant Classification Scheme 2023: The p.Y122C variant (also known as c.365A>G), located in coding exon 2 of the TERT gene, results from an A to G substitution at nucleotide position 365. The tyrosine at codon 122 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
