This alteration results from a G to A substitution at nucleotide position 1825, causing the aspartic acid (D) at amino acid position 609 to be replaced by an asparagine (N). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This variant was reported in individual(s) with features consistent with ATP1A3-related neurologic disorders (Wu, 2022; Wu, 2023). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
