This variant lies in the NOTCH1 gene (transcript NM_017617.5) at coding-DNA position 2898 through coding-DNA position 2899, replacing the reference sequence with GA. Submitter rationale: The c.2898_2899delCTinsGA variant, located in coding exon 18 of the NOTCH1 gene, results from an in-frame deletion of CT and insertion of GA at nucleotide positions 2898 to 2899. This results in the substitution of serine and tyrosine residues for arginine and asparagine residues at codons 966 to 967. This amino acid region is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
