Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000890.5(KCNJ5):c.902T>A (p.Val301Asp), citing Ambry Variant Classification Scheme 2023: The p.V301D variant (also known as c.902T>A), located in coding exon 1 of the KCNJ5 gene, results from a T to A substitution at nucleotide position 902. The valine at codon 301 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
