NM_181486.4(TBX5):c.841C>G (p.Leu281Val) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L281V variant (also known as c.841C>G), located in coding exon 7 of the TBX5 gene, results from a C to G substitution at nucleotide position 841. The leucine at codon 281 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
