Uncertain significance for Retinitis pigmentosa 71; Short-rib thoracic dysplasia 10 with or without polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015662.3(IFT172):c.3196C>G (p.Arg1066Gly), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the IFT172 gene (transcript NM_015662.3) at coding-DNA position 3196, where C is replaced by G; at the protein level this means replaces arginine at residue 1066 with glycine — a missense variant. Submitter rationale: This sequence change replaces arginine, which is basic and polar, with glycine, which is neutral and non-polar, at codon 1066 of the IFT172 protein (p.Arg1066Gly). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with IFT172-related conditions. ClinVar contains an entry for this variant (Variation ID: 1019702). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
