Uncertain significance for Majeed syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001375808.2(LPIN2):c.191T>C (p.Val64Ala), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with LPIN2-related conditions. This variant is present in population databases (rs754730690, ExAC 0.001%). This sequence change replaces valine with alanine at codon 64 of the LPIN2 protein (p.Val64Ala). The valine residue is highly conserved and there is a small physicochemical difference between valine and alanine.
